Can someone be mekyem the mitzvah of eradicating amalek by writing amalek on paper and then erasing or crossing out or destroying the name? If so must it be on a real klaf?

No, the mitzvah is only fulfilled by means of actually destroying Amalek (the nation), and erasing from kelaf is not a fulfillment of the mitzvah. Writing it on kelaf will not help for this. It may, however, serve as a reminder of the obligation to wipe out Amalek, which is a constant mitzvah, and which is performed by the reading of Parashas Zachor. For this purpose, kelaf is not required.
