Hello,
If I have a bag of sand that I was using to hold down a door in an unusual manner how would I make that bag not muktzah do I need to designate it with speech or do an action to it or can I use it for that purpose a couple of times before Shabbos or even once and where are these halachos discussed.

If I am understanding your question correctly, you already used the sand bag to hold a door open or closed during the week, and you want to know if it will be muktza to use it on Shabbos. Being that you already used it during the week, the bag is now considered designated for your use, and it isn’t muktza. Therefore, there is no need to do anything to it.
