There are two issues here, taking the pill regarding taking medications on Shabbos, Yom Tov or Yom Kippur. For that do whatever you do every week. As far as Yom Kippur is concerned, if you take the pill without any water, you wouldn’t have any issue with that. If you can’t take it without a little bit of water, the water should be made bitter so you won’t feel benefit from it. If the pill is sweet, it should be wrapped in a thin tissue, so it won’t be considered eating.
